01 — PROCESS

Where does each step fit?

Define the intended use and conditions to identify the required role of each item.

01

Filling and fitting the protective support

Apply the approved fill volume and headspace, and arrange the bag, ports and tubing within the protective support.

02

Freezing, storage and transport

Manage product temperature, loading position and criteria for completion of freezing, and apply validated packaging and transport conditions.

03

Thawing and quality verification

Define the thawing endpoint and remixing conditions, and check for leaks and required quality attributes such as concentration, aggregation and activity.

02 — SELECTION

What should be selected and verified?

Select the bag and protective support with consideration for expansion during freezing, low-temperature handling and transport impacts. Evaluate the actual product temperature history and quality after thawing, rather than defining the process from the freezer setpoint alone.

Volume or film name alone is insufficient. Review the actual fluid, contact conditions, equipment compatibility and supplier evidence.

03 — OPERATING CHECKS

Use operating data to identify improvements.

Distinguish setpoint from product temperature

Even in the same freezer, freezing rate differs with bag size and loading position. Define product temperature at representative locations and process endpoint criteria.

Do not judge quality from low-temperature resistance alone

Protein aggregation or changes in activity may occur even when the bag withstands the conditions. Compare quality after freezing and thawing using the actual drug substance composition and concentration.

Protect the ports of frozen bags

Do not forcefully bend or subject the bag and tubing to impact at low temperatures. Review protective supports, transport packaging and operating procedures together.

04 — QUESTIONS

Frequently asked questions

Can all freezing bags be placed in liquid nitrogen?

No. Freeze–thaw bags for bulk drug substance and cryopreservation bags for cells may differ in intended use and permitted conditions. Confirm the target temperature and storage method in the product specifications.

If a small sample is stable, will a large bag perform the same way?

Volume, thickness, loading and heat-transfer conditions differ. Testing and quality evaluation that reflect the temperature history at production scale are required.
